Molecular interactions of ErbB receptor tyrosine kinases and integrin β1: implications for tumor therapy

We set out to investigate the molecular interactions of ErbB receptor tyrosine kinase proteins and integrin molecules to shed light on their possible interfering, correlative roles behind trastuzumab resistance of breast cancers and radioresistance of astrocytomas. To study the contingent mutual interplay of neighbouring receptors, we have implemented a method, two-sided FRET (tsFRET), for the investigation of the relationship of associations of two molecule pairs of three arbitrarily chosen molecular species. For the evaluation of individual differences of clinical samples, we adapted the abFRET method for quantitatively assessing in situ molecular interactions on intraoperative fresh frozen tissue sections. According to our results, it is tempting to speculate that the homoassociation state of ErbB2 is dynamically modulated by the interaction with integrin β1 on breast cancer cells in vitro, although a strong correlation toward trastuzumab resistance could not be affirmed on the basis of our study. It may, however, be assumed that higher integrin β1expression alters the signal transduction of breast cancer cells, thereby presenting an alternative pathway for the activation of the MAPK and PI-3K/Akt routes and giving rise to the metastasizing ability. As our preliminary experiments confirmed dynamically modifiable ErbB-integrin β1 interactions, and a protein level screen on astrocytoma cells proved integrin β1 overexpressed along with ErbB1, we aimed to assess their possible interaction behind gliomagenesis. Two cellular model systems showing expression profiles similar to low and high grade astrocytoma tumors have been generated and we showed that increased ErbB1 expression was consequently followed by the presence of excess integrin β1 in the membrane. Decreasing radiosensitivity correlated with increasing ErbB1 and integrin β1 expression levels, and also with decreasing ErbB1 homoassociation and increasing ErbB1 – integrin β1 heteroassociation rates. Radioresistance could be explained by the increased basal Akt phosphorylation, and augmented output from the PI-3K/Akt pathway. After EGF stimulation in the sublines with higher ErbB1 and integrin β1 expression this increased radioresistance could be reverted by inhibition of PI-3K. The enhanced ErbB1 - integrin β1 interaction was also detectable in grade IV versus grade II astrocytoma fresh frozen sections and appeared to be an efficient predictor of patient survival and disease relaps. The abFRET procedure proved to be a useful, cell function-related diagnostic method in assessing in situ ErbB1–integrin β1 heteroassociation, which implies potential exploitation of similar investigations that may allow for discernment of prognostically diverse patient subgroups warranting the application of certain single or combined therapeutic modalities.

Kutatásunk az ErbB receptorok és integrinek molekuláris interakcióinak vizsgálatára irányult, hogy felfedjük kölcsönhatásaik potenciális szerepét az emlőtumorok trastuzumab rezisztenciájának és az astrocytomák sugárrezisztenciájának hátterében. A szomszédos membrán fehérjék kölcsönös interakcióinak vizsgálatára egy új módszert - kétoldali FRET - fejlesztettünk ki, amely alkalmas három tetszőlegesen megválasztott molekula által képzett két különböző molekuláris interakció korrelációjának mérésére. Egyedi klinikai tumorokban fennálló és esetenként különböző molekuláris kölcsönhatások pontos feltérképezése céljából pedig in situ friss fagyasztott metszetekre adaptáltuk az abFRET módszert. Eredményeink alapján felmerül, hogy emlőtumorokban az ErbB2 homoasszociációját in vitro dinamikusan modulálja az ErbB2-β1 integrin heteroasszociáció, azonban a trastuzumab rezisztencia irányában egyértelmű összefüggés nem alapozható tanulmányunkra. Mindazonáltal feltételezhető, hogy a fokozott β1 integrin expresszió befolyással van az emlőtumor sejtek jelátvitelére és mintegy alternatív utat biztosít a MAPK és PI-3K/Akt útvonalak aktiválódása, illetve a metasztatizáló képesség potenciális kifejlődése számára. Miután előzetes vizsgálataink bizonyították az ErbB2 receptor és a β1 integrin molekula dinamikus kölcsönhatását és astrocytoma sejtvonalakon végzett szűrővizsgálataink a már jól ismert emelkedett ErbB1 expresszió mellett a β1 integrin fokozott kifejeződését igazolták, célul tűztük ki az ErbB1-β1 integrin interakció vizsgálatát a gliomagenezis hátterében. Két sejtes modell rendszert állítottunk fel, amelyek az alacsony és magas grádusú astrocytomákhoz hasonló ErbB1 és β1 integrin expressziós mintázattal rendelkeztek. Igazoltuk, hogy a fokozott ErbB1 expresszió a β1 integrin expresszió növekedésével és a sugárérzékenység csökkenésével társult. Ezzel párhuzamosan az ErbB1 homoasszociáció csökkenését az ErbB1-β1 integrin heteroasszociáció fokozódása követte. A sugárrezisztenciát magyarázhatta az ezzel együtt fokozódó bazális Akt foszforiláció és PI-3K/Akt szignalizáció. EGF kezelés hatására a PI-3K útvonalon bekövetkező augmentáció a PI-3K gátlásával visszafordítható volt, amely egyúttal a fokozott sugárrezisztencia elvesztésével is együtt járt. Friss fagyasztott II. és IV. grádusú astrocytoma mintákon szintén erős ErbB1-β1 integrin interakciót mutattunk ki, amely a túlélés és a relapszus hatékony prediktorának bizonyult. Az ErbB1-β1 integrin heteroasszociáció in situ vizsgálatában az abFRET módszer diagnosztikus jelentőségű módszernek bizonyult a sejtfunkciós változások viszonylatában, és felveti a lehetőségét hasonló tanulmányok elvégzésének, amely hozzájárulhat a betegek prognosztikai csoportosításához és egyedi terápiás megközelítések alkalmazásához.
