A Melitaea genus Mellicta subgenusába tarozó lepkefajok populációi Európa egyes részein hanyatlóak vagy alacsony egyedszámúak, így konzervációbiológiai és természetvédelmi jelentőségük igen nagy. A subgenus taxonómiailag bonyolult, mivel külső jellegeik igen variábilisak, valamint rejtett fajok, alfajok lehetnek jelen. Az élőhelyeik nagyon hasonlóak, bizonyos élőhelyeken egymás mellett fordulnak elő. Életmenet stratégiájukból adódóan a Melitaea-fajok populációiban nagymértékű lehet a genetikai sodródás a véletlenszerű események (sztochasztikus hatások) bekövetkezése miatt. Ugyanis egy nőstény egy csomóba viszonylag nagy mennyiségű petét rak le, és a fiatal hernyók is egy darabig társasan élnek, ezért egy nőstény összes utóda egyszerre van kitéve a különböző környezeti faktorok hatásainak. Az általam vizsgált három tarkalepkefaj hasonló fejlődésmenetű, de életciklusukban egy igen lényeges eltérést lehet megfigyelni, ez pedig a voltinizmus. Míg a M. britomartis és a M. aurelia évente egy nemzedékes, addig a M. athalia esetében két típust lehet elkülöníteni. Az egyik fenológiai típusnak egy generációja van egy évben (monovoltin), míg a másik típusnak kettő (bivoltin). Továbbá a M. athalia lárvális fejlődése kis mértékben eltér a másik két fajétól, ugyanis hernyóik már a diapauza előtt szétszélednek a hernyófészekből, egyedileg táplálkoznak és egyedileg esztiválnak illetve hibernálnak. Ezek a lárvák a másik két faj hernyóinál jóval szélesebb tápnövényspektrumot képesek hasznosítani. Vizsgálataim során a Mellicta subgenus három fajának Kárpát-medencei populációiban tanulmányoztam a genetikai variabilitás szerkezetét, továbbá a differenciálódás mértékét. Ennek a vizsgálatnak az eredményeire alapozva különös figyelmet fordítottam a M. athalia két fenológiai típusának vizsgálatára, valamint a fajon belüli differenciálódás mintázatának feltárására. Ezért ennek a fajnak külön vizsgáltam a genetikai variabilitását és fajon belüli differenciálódásának mértékét is, továbbá párhuzamos elemzést végeztem a fenotípusos variabilitás és differenciálódás vonatkozásában. Az eredményeket felhasználva a genetikai és a fenotípusos variabilitás szintje illetve struktúrája közötti korrelációt is megvizsgáltam a M. athalia populációkban.

Dolgozatom főbb eredményeit az alábbiakban foglalom össze: A vizsgált három Melitaea-faj enzimpolimorfizmus vizsgálata során három Evolúciósan Szignifikáns Egységet (ESU-t) különítettem el a Mellicta subgenuson belül a három fajnak megfelelően. Az eredményeim alapján a M. britomartis és a M. aurelia közelebbi, míg a M. athalia távolabbi rokonnak bizonyultak. Ez ellentmond Wahlberg és mtsai. eredményeinek, mivel a vizsgálataik alapján a M. athalia és a M. britomartis bizonyult közeli rokonnak. Természetesen azt meg kell jegyezni, hogy ők más módszerrel, a Melitaeini tribusz összes fajának egy-egy egyedét vizsgálták. A M. athalia két fenológiai típusa között egyik analízis során sem mutatkozott különbség, azaz nem tekinthetők genetikailag két külön csoportnak. Mindezek alapján valószínűsíthető, hogy az egy illetve két generációs jelleg egyfajta fenotípusos plaszticitás, vagyis az elkülönülés élőhelybeli sajátosságokon alapszik. Az analízisek eredményei azonban további kérdéseket vetettek fel, ugyanis a M. athalia populációk genetikailag nagymértékű differenciálódást mutattak egymástól. A M. athalia genetikai valamint morfometriai adatainak párhuzamos elemzése során fényt derítettem a populációk differenciálódási struktúrájának mintázatára. Minden genetikai analízisben kimutattam a populációk regionális differenciálódását, ráadásul ebben az esetben a régiók két jól elkülöníthető csoportot alkottak a keleti-, ill. nyugati Kárpát-medencének megfelelően. A M. athalia két fenológiai típusa ebben a vizsgálatban sem különült el egymástól egyik analízisben sem, tehát ebben a típusú vizsgálatban is azt tapasztaltam, hogy nincs a két csoport között genetikai különbség. A fenotípusos differenciálódást megvizsgálva külön-külön a szárny valamint a hímivarszerv jellegeire vonatkoztatva szintén a populációk regionális differenciálódását tapasztaltam. A régiók a szárnyjellegek vonatkozásában részlegesen különültek el a keleti-, ill. nyugati Kárpát-medencének megfelelően, azonban az ivarszerv jellegei esetében ilyen mintázat nem volt megfigyelhető. Eredményeim azt tükrözik tehát, hogy a genetikai valamint a morfometriai vizsgálatok eredményei hasonlóak, ugyanis mindkét esetben a populációk regionális differenciálódása volt kimutatható. Ez a mintázat azonban az enzimpolimorfizmus vizsgálatok során markánsabbnak bizonyult, mivel ebben az esetben a régiókra ráépül egy magasabb hierarchia szint is, amely révén a földrajzi régiók a keleti-, ill nyugati Kárpát-medencének megfelelően csoportosulnak. Bár a mintavételi stratégiából adódóan nem zárható ki a genetikai klin megléte, azonban az eredmények sokkal erősebben támasztják alá a keleti-, ill. nyugati Kárpát-medence populációinak elkülönülését. A genetikai variabilitást megvizsgálva a Mellicta subgenuson belül mindhárom faj tekintetében magas szintű enzimpolimorfizmust tapasztaltam. A vizsgált 15 enzimlokusz közül mind rendelkezett alternatív allélokkal, sőt a legtöbb igen polimorfnak bizonyult. A polimorfizmus mutatói mindhárom esetben magasnak mutatkoztak, de a három faj közül M. athalia volt a legvariábilisabb. Mivel a Melitaea-fajok az r-K kontinuum r-hez közelebbi végén helyezkednek el, a populációikat jelentős mértékben érintik a sztochasztikus hatások, így intenzív drift várható melynek következményeként általánosságban a populációknak alacsony szintű lesz a variabilitása. A dolgozatom eredményei ezzel ellentétben magas szintű variabilitást tükröztek, tehát nem a drift tűnt a variabilitás szintjét befolyásoló elsődleges tényezőnek. Két további lehetséges tényező hatását elemezve (általános ökológiai jellemzők eltérései, különös tekintettel a tápnövény hasznosítás spektrumának nagyságára, vizsgált populációk helyzete a faj areáján belül) oly módon, hogy a 3 Mellicta faj variabilitását a Melitaeini tribusz 4 további fajának (Euphydryas maturna, E. aurinia, Melitaea phoebe és M. ornata) enzimpolimorfizmu adataival hasonlítottam össze. Az eredmények azt mutatták, hogy a vizsgált fajok variabilitásának mintázatát a centrális–marginális hipotézis magyarázza meg a legteljesebben. A morfológiai jellegek változatossága sok vizsgálatban tükrözte a genetikai variabilitás szintjét és szerkezetét. Meglepő módon a Levene-változók százalékos értékei azt sugallták, hogy az ivarszerveken mért jellegek variabilitása magasabb, mint a szárnyjellegeké. Ez az eredmény váratlan, hiszen a szárnyak köztudottan ki vannak téve a környezeti tényezők hatásainak, míg az ivarszervek morfológiája feltehetően sokkal jobban behatárolt. A M. athalia fenotípusos variabilitásának vizsgálata során a különböző analízisek arra világítottak rá, hogy sem a szárny, sem pedig az ivarszerv jellegek variabilitásában nincs egyértelmű, szignifikáns különbség a fenotípusos variancia mértékében a hierarchia felső szintjén. Ugyanakkor a két jellegcsoport esetében ellentétes tendenciát tapasztaltam a morfometriai variabilitás mértékében a Kárpát-medence keleti, illetve nyugati területének populációi között. Míg a szárnyakon mért jellegek varianciája párhuzamos tendenciákat mutat a genetikai varianciával, addig az ivarszervek jellegeinél ellentétes trend volt megfigyelhető. A genetikai és a fenotípusos variabilitás szintje közötti korrelációs vizsgálat eredményei első megközelítésben azt mutatták, hogy a genetikai és morfológiai variabilitás között nincs szoros kapcsolat sem a szárnyak, sem pedig az ivarszerv jellegeit tekintve. A részletesebb elemzés azonban arra világított rá, hogy a szárnyjellegek két típusának (méret+alak és mintázat) variabilitása között nincs korreláció. Így ezt a két jellegcsoportot külön kezelve a vizsgálati eredmények azt tükrözték, hogy a kevésbé variábilis szárnyméret és alak jobban jelzi a genetikai variabilitás szintjét, mint a szárny mintázata, melyről számottevő mértékű plaszticitást tételezhetünk fel. Az a tény, hogy a méretet és alakot meghatározó jellegek fenotípusos varianciája alacsonyabb szintű, mint a mintázatot jellemzőké, valamint hogy a szárnyméret varianciája szignifikáns korrelációban van az enzimpolimorfizmus mértékével azt sugallja, hogy ezeknek a jellegeknek a fenotípusos varianciáját nagyrészt genetikai faktorok határozzák meg. A genetikai és a fenotípusos differenciálódás mértékének összehasonlítása során végzett Mantel teszt eredményei azt mutatták, hogy a szárny jellegeit tekintve szoros korreláció van a differenciálódás ezen két szintje között. Ugyanakkor érdekes, hogy a variabilitás szintjéhez hasonlóan a differenciálódás mértékében sem tapasztaltam korrelációt a genitália jellegei és az enzimpolimorfizmus között. Ráadásul a két morfometriai jellegcsoport (szárny-, és genitáliajellegek) differenciálódása sem korrelált egymással. A korreláció hiánya arra utal, hogy eltérő környezeti tényezők hatnak a két morfológiai jellegcsoport fenotípusos varianciájára.

Species of the Mellicta subgenus are listed as vulnerable or declining in several countries of Europe. Therefore, these species are of obviously great conservation concern. The subgenus is taxonomically complicated due to the great variety of the species in their external characters. Even the existence of cryptic species has been suggested. The habitat requirements and food plants of Melitaea species partly overlap, they often co-occur within the same habitat. Melitaea species have fairly similar life cycle: females lay a great number of eggs in a few batches and their caterpillars remain together feeding gregariously. As a consequence, all offspring of a female might be affected by various environmental factors. Due to this special reproductive strategy populations of the Melitaea species are greatly exposed to stochastic events and genetic drift is likely to have a large effect on their genetic composition. The studied three Melitaea species differ in voltinism. Hungarian populations of M. britomartis and M. aurelia have a single brood per year, while M. athalia has two ecotypes: one has one brood per year (monovoltine) and the other usually has two (bivoltine). Moreover larval development of M. athalia is slightly different compared to the other two. Caterpillars of this species spread before the diapause, feed and aestivate/hibernate solitarily. Furthermore, larvae of M. athalia can exploit a wider range of food plants than those of the other two Melitaea species. In my surveys, three Melitaea species were studied and I have analysed the structure of genetic variation and the level of differentiation of these species in the Carpathian Basin. I was particularly interested in the level of differentiation among M. athalia populations with special regard to the two ecotypes of this species. Accordingly, I have analysed the pattern of genetic differentiation among M. athalia populations in the Carpathian Basin. In addition to the study of enzyme polymorphism I also surveyed the structure of morphometric variation using selected characteristics of wings and external genitalia. Thus, the structure of genetic and phenotypic variation was investigated in parallel. This approach allowed me to analyse the association between genetic and phenotypic variation in this species.

The results can be summarized in the following points: Three distinct Evolutionary Significant Units (ESUs) were detected within the Mellicta subgenus in the Carpathian Basin corresponding to the three species. The results revealed that M. britomartis and M. aurelia seem to be closely related, while M. athalia appears to be a further relative. This result contradicts to those of Wahlberg and his coworkers’ surveys. In their tree, M. athalia and M. britomartis were closely related, while M. aurelia occurred in a neighbouring branch with other related species. It has to be mentioned, however, that their approach was entirely different: they used other markers to study the phylogeny of all species of the Melitaeini tribe using 1-3 individuals from each. None of my results supported the differentiation between the two ecotypes of M. athalia, that is, they cannot be considered as genetically separated groups. The apparent difference in the life cycle between the mono- and bivoltine ecotypes is probably the consequence of the high phenotypic plasticity of the species and it is probably the consequence of habitat quality. The results also indicated that M. athalia populations were highly differentiated from each other. In parallel analyses of genetic and morphometric data I was able to describe the pattern of differentiation in M. athalia. All genetic analyses suggested a clear regional differentiation. Moreover, the regions could be separated to the East and West geographic zones of the Carpathian Basin. The two ecotypes of M. athalia were not separated in these analyses either implying that there are no genetic differences between them. Findings of the morphometric studies of wings and genital organs showed a regional pattern of differentiation of the phenotypic variation as well. Considering the characters measured on the wings, the separation between the regions of the East and West geographic zones of the Carpathian Basin was less clear than in the study of enzyme polymorphism. This West-East separation, however, was not detected in the characters measured on the genital structures. Accordingly, the results of my work indicated that the structure of genetic and phenotypic variation exhibited certain similarities in M. athalia, as regional pattern of differentiation was detected for both types of variation. The geographic pattern, however, proved to be more complex in the genetic analyses as the regions composed two clear groups corresponding to the East and West geographic zones of the Carpathian Basin. Considering our sampling strategy the presence of a genetic cline cannot be completely ruled out. Nevertheless, the results support the West-East separation of the populations more strongly. Analysing the level of genetic variability in the Mellicta subgenus, high enzyme polymorphism was found in all three species. All studied 15 enzyme loci had alternative alleles; moreover most of them were highly polymorphic. The classidcal parameters of genetic variation were high in all three species, but the M. athalia seemed to be the most divers. Since Melitaea species take place at the r side of the r-K continuum, stochastic events affect their populations to a great extent. Thus, genetic drift was expected resulting in low level of variability within the populations. As opposed to my expectations the results indicated an extremely high level of enzyme polymorphism. Looking for the explanation of this high variation the effect of two further possible factors was considered: (i) general ecology of the species especially the range of host plant utilization; (ii) the site of the studied populations relative to the distribution area of the species. Moreover, the genetic data of the three Mellicta species were compared to those of four other species of the Melitaeini tribus (Euphydryas maturna, E. aurinia, Melitaea phoebe and M. ornata). The results showed that the pattern of genetic variation in the seven species was best explained by the central-marginal hypothesis. In the analysis of phenotypic variation first we compared the different morphometric characters. Surprisingly, the Levene-variables expressed as percentages of trait averages suggested that the variation of characters of external genitalia were higher than those of the wing traits. This result was unexpected, as it is generally known that wing traits are more affected by environmental factors, while the morphology of external genitalia is more determined by genetic factors. In many surveys, the genetic structure of populations was reflected in the variability of morphological characters. Thus, we contrasted the phenotypic and genetic variation in the M. athalia populations studied. Both types of variation exhibited a geographic pattern; that is a more or less expressed regional differentiation was observed in the analysis of enzyme polymorphism and morphometric traits of the wings and external genitalia as well. Few significant differences were found in the phenotypic variation of the characters of the wings and genitalia between either the two ecotypes or the two zones of the Carpathian Basin. Nevertheless a tendency was found for several traits of the wings for higher variation in the populations of the East zone of the Carpathian Basin. This tendency was in accordance with that observed for enzyme polymorphism. In contrast, the traits of the genital organs showed an opposite tendency; i.e. the populations of the West zone of the Carpathian Basin seemed to be more variable than those of the East one. Testing the correlation between the level of phenotypic and genetic variation at first sight I did not detect significant relationship between them neither for the traits of external genitalia nor for the wing characters. In a more detailed analysis, however, I found that the two types of wing traits (size+shape and pattern) are not correlated with each other. Accordingly, I handled the two types of wing characters separately in all further analyses, and found that variation in the size and shape of the wings which are less variable and less influenced by environmental factors is significantly correlated to the level of enzyme polymorphism. That is, phenotypic variation of these characters is a better indicator of genetic variation than that of wing pattern which is assumed to be more plastic. These results suggested that the phenotypic variance of the size and shape of the wings was mainly determined by genetic factors. Nevertheless, the level of morphometric variation of the traits of genitalia was not correlated to either the amount of enzyme polymorphism or to that of phenotypic variation in the size and shape of the wings. I have used a series of Mantel tests to compare the level of genetic and morphometric differentiation among the M. athalia populations. The results indicated a relatively strong correlation between phenotypic differentiation estimated on the basis of the size and shape characters of the wings and genetic differentiation characterised by enzyme polymorphism. No correlation was found, however, between the level of genetic and phenotypic differentiation when the traits of external genitalia were used in the analyses. Moreover, phenotypic differentiation was not correlated either between the two types of morfometric traits. The lack of correlation between wing traits and those of the genitalia implies that different environmental factors influence them.
