A nervus acusticus akcióspotenciál-mintázata a hang valamennyi érzékelhető tulajdonságát hordozza. A hallóideg aktivitásának hangérzetté való alakításáért egy olyan központi idegrendszeri hálózat felelős, ami a nucleus cochlearisszal kezdődik és a hallókéregben ér véget. A nucleus cochlearis azonban nem pusztán a központi idegrendszeri hallópályák kiinduló pontja, de ez a struktúra felelős azért is, hogy a beérkező hallási információ több, egymással párhuzamosan haladó, felszálló pályára kerüljön át. Ezen felszálló pályák egy jelentős hányada a hang egy-egy speciális tulajdonságáról közvetít információt, ami nélkülözhetetlen a hanginger jellemzőinek központi idegrendszeri rekonstrukciójához. A specifikus és intenzív kutatómunka ellenére is hézagosak az ismereteink a nucleus cochlearis és az abban található nagy számú neurontípus funkciójával kapcsolatosan. Különösen kevés adat áll rendelkezésre a mag legkisebb, ugyanakkor legnagyobb számban előforduló sejttípusáról, a szemcsesejtekről. Még ennél is kevesebbet tudunk az elsősorban a nucleus cochlearis dorsalis részében előforduló, igen jellegzetes morfológiát mutató sejttípusról, a Purkinje-szerű sejtekről (PLC). Utóbbi neuronféleség esetében még az sem ismert, hogy vajon részét képezik-e a nucleus cochlearis neuronhálózatának, vagy funkció nélküli, ectopiás Purkinje-neuronokról van szó. Jelen disszertáció patkány nucleus cochlearis dorsalis szemcse- és Purkinje-szerű sejtjeinek vizsgálatát tűzte ki célul. Kísérleteinket elektrofiziológiai és képalkotó módszerek kombinált alkalmazásával végeztük. Munkánk első részében a Purkinje-szerű sejtek morfológiai jellemzőit vizsgáltuk, amihez ezen neurontípus intenzív calbindin-pozitivitása nyújtott segítséget. Kettős immunjelölés és konfokális mikroszkópia alkalmazása után megmutattuk, hogy a PLC-k rendelkeznek a GABA bioszintéziséhez nélkülözhetetlen glutamát dekarboxilázzal. Ez a megfigyelés azt valószínűsíti, hogy a PLC-k GABAerg gátlóneuronok. Túlélő agyszelet-preparátum és teljes-sejtes patch-clamp segítségével az irodalomban elsőként írtuk le ezen sejttípus akcióspotenciál-tüzelési mintázatát. Bebizonyítottuk továbbá, hogy a Purkinje-szerű sejtek rendelkeznek a hiperpolarizáció hatására aktiválódó, nem-specifikus kationárammal (Ih). A PLC-k sejttestéről elvezetett posztisznaptikus áramok vizsgálata arra is bizonyítékot szolgáltatott, hogy a sejttípus aktív részese a DCN neuronhálózatának. Megállapítottuk, hogy a PLC-kről elvezethető gátló hatású posztszinaptikus áramok 76%-a glicinerg szinapszisok aktivitását tükrözi, míg a fennmaradó 24% GABAerg bemeneteken keresztül érkezik. Arra is rámutattunk, hogy bár a cerebellaris Purkinje-sejtekhez hasonlóan a PLC-k is erős cerebellin-1 immunpozitivitást mutatnak, a fehérje sejten belüli eloszlása különbözik a két sejtféleségben. Az egyik olyan neurontípus, aminek aktivitása jelentősen módosíthatja a nucleus cochlearis egyéb sejtjeinek – beleértve a PLC-ket is – aktivitását és működését, a cochlearis szemcsesejt. A szemcsesejtek különös jelentőségét az adja, hogy somatosensoros információ is eléri őket. Ebből kiindulva megalapozottnak tűnik az a feltételezés, hogy a szemcsesejtek alapvető jelentőségűek a somaticus és acusticus stimulusok nucleus cochlearison belüli ötvözésében. Bár léteztek arra adatok, hogy a szemcsesejtek működését kolinerg moduláció is befolyásolhatja, a jelenség részleteinek leírására ezidáig nem került sor. Ebből kiindulva munkánk második felében azt vizsgáltuk, hogy hogyan befolyásolja a szemcsesejtek intracelluláris kalciumkoncentrációját a kolinerg stimuláció. Kísérleteinket túlélő agyszeletekben elhelyezkedő, kalciumérzékeny fluoreszcens festékkel feltöltött szemcsejteken végeztük. A vizsgálatsorozat kezdetén bizonyítottuk a szemcsesejtek gyors típusú intracellularis Ca2+-jeleinek akcióspotenciál-kapcsolt voltát, majd kidolgoztunk egy módszert ezen Ca2+-jelek megbízható, automatikus azonosítására és kinetikai paramétereik jellemzésére. Megmutattuk, hogy a kolinerg stimuláció még a serkentő és gátló hatású szinaptikus transzmisszió egyidejű gátlása esetén is szignifikánsan megnöveli a szemcsesejtek aktivitását, ezáltal az általuk produkált gyors típusú Ca2+-jelek frekvenciáját. Specifikus antagonistákkal végzett kísérletekben arra is rámutattunk, hogy az általunk feltárt kolinerg hatást muszkarinos receptorok közvetítik. Bebizonyítottuk, hogy bár a szemcsesejtek mind az M1-, mind az M3-típusú muszkarinos receptort expresszálják, a funkcionális hatások kialakításáért döntően az utóbbi receptortípus aktiválódása felelős. Eredményeink hozzásegítenek annak megértéséhez, hogy a szemcse- és Purkinje-szerű sejtek hogyan illeszkednek a nucleus cochlearist felépítő neuronális hálózatba, így pontosabb képet nyerhetünk ezen bonyolult, a hallási információ központi idegrendszeri feldolgozásához nélkülözhetetlen struktúra működéséről.

The action potential firing pattern of the acoustic nerve encodes all recognisable features of the sound. The conversion of the activity of the auditory nerve to acoustic sensation is carried out by a neuronal network that begins in the cochlear nucleus and terminates in the auditory cortex. The cochlear nucleus, however, is not the mere starting point of the central auditory pathways but it passes the incoming acoustic information onto several, parallel ascending pathways. A number of these pathways relay information about specific features of the sound. This arrangement is essential for the reconstruction of the overall characteristics of the auditory stimuli. This highly complicated task is carried out by the central auditory system. Despite all the intense and specific research efforts, our knowledge about the function of either the cochlear nucleus or its individual cell types is still obscure. Particularly little information is available about the granule cells that represent the smallest, yet most numerous cell type of the cochlear nucleus. Even less is known about the Purkinje-like cells (PLCs). PLCs are mostly located in the dorsal cochlear nucleus and show highly characteristic morphology. It is still not clear, for example, whether PLCs are integrant parts of the neuronal network of the cochlear nucleus or they are mere ectopic Purkinje neurons, without function. For these reasons, the present work aimed at investigating the granule neurones and the PLCs of the rat dorsal cochlear nucleus. The experiments have been carried out by combined application of electrophysiological and imaging techniques. In the first part of our work the morphological characteristics of the PLCs were investigated. This phase of our experiments exploited the intense calbindin-specific immunopositivity of the PLCs. Using double immunolabelling and confocal microscopy; we demonstrated that PLCs express glutamate-decarboxylase, an enzyme which is essential for GABA biosynthesis. This observation suggests that the PLCs are GABAergic inhibitory neurones. Using whole-cell patch-clamp in a brain slice preparation, for the first time in the literature, we revealed the action potential firing pattern of the PLCs. We showed that PLCs possess the hyperpolarisation-activated non-specific cationic current (Ih). Analysis of postsynaptic currents recorded from the somata of PLCs indicated that this cell type is an active member of the neuronal network situated in the dorsal cochlear nucleus. We showed that 76% of the inhibitory postsynaptic currents recorded from the PLCs reflect the activity of glycinergic synapses, while the remaining 24% reach the PLC bodies via GABAergic inputs. We also pointed out that although cerebellar Purkinje-neurones and PLCs are similar in terms of their strong cerebellin-1 immunopositivity, the intracellular distribution of cerebellin-1 is different in these cell types. One of the neurones whose activity may significantly modify the function of all other cell types of the dorsal cochlear nucleus — including PLCs — is the cochlear granule cell. Granule neurones are particularly important because they receive somatosensory information, too. It is reasonable to assume, therefore, that granule cells play fundamental roles in the integration of auditory and somatic stimuli within the cochlear nucleus. Although there have been data indicating that the function of the granule cells is affected by cholinergic modulation, a detailed analysis of this mechanism is yet to be performed. Consequently, in the second part of the present work, we have been studying the effects of cholinergic stimulation on the intracellular calcium concentration of the granule neurones. The experiments were carried out on calcium-sensitive fluorescent dye-loaded granule cells situated in a thin slice preparation. At the beginning of the work, we demonstrated the action potential-coupled character of the rapid calcium transients recorded from the granule cells. In the next step we introduced a method that automatically detects calcium concentration changes and determines their kinetic features. We showed that cholinergic stimulation significantly increases the activity of the granule cells and, consequently, the frequency of their rapid calcium transients — even when both excitatory and inhibitory synaptic inputs are blocked. Using specific antagonists, we demonstrated that this cholinergic influence is mediated by muscarinic receptors. We showed that although granule neurones express both M1 and M3 muscarinic receptors, the functional effects are primarily mediated by the activation of M3 receptors. Our results help us to understand better how granule neurones and PLCs are integrated into the complex neuronal network of the cochlear nucleus. Consequently, as the result of our observations, one may have more comprehensive information depicting the function of this highly complicated structure without which central processing of the auditory information would be impossible.
