Glikopeptid antibiotikumok szintetikus módosításai

Doktori kutatómunkám során két glikopeptid antibiotikum, a teikoplanin és a vankomicin félszintetikus módosításait végeztem el abból a célból, hogy az anyavegyületekre rezisztens, főleg Gram-pozitív kórokozók ellen hatásos származékokat, illetve antivirális hatású vegyületeket nyerjek. A teikoplanin A3-2 pszeudoaglikon (TC) előállítására egyszerű, költséghatékony eljárást dolgoztam ki, majd e vegyület N-terminálisához pl. réz(I)-katalizált azid-alkin click reakció (CuAAC), N-szulfonilezés vagy guanidincsoport kialakításának segítségével elsősorban lipofil karakterű oldalláncok hozzákapcsolását végeztem el. A kapott származékok jelentős része az anyavegyületeknél jóval magasabb aktivitást mutatott a glikopeptid rezisztens baktériumtörzsek ellen. Néhány vegyület antivirális hatással is rendelkezett, pl. influenzavírusok, ill. koronavírusok ellen. Az egyes oldalláncok szerkezetének szisztematikus módosításával az antibakteriális és az antivirális hatás is hangolhatónak bizonyult, valamint a citotoxicitás csökkentése is sikeres volt néhány esetben. Az N-terminális módosítása mellett peptidkapcsolással a C-terminálison bázikus aminocsoporttal szubsztituált amidcsoportot kialakítva az antibakteriális hatást esetenként sikerült tovább növelnem. Szelektív (antibakteriális hatással nem rendelkező) antivirális vegyületeket sikerült előállítanom az influenzavírusok ellen legaktívabb és legkevésbé toxikus teikoplanin származékok oldalláncainak a vankomicin aglikon hexapeptid (VAHP) N-, illetve C-terminálisához kapcsolásával. Gram-negatív baktériumok elleni hatás elérése céljából a VAHP újraacilezésével, majd aminoetilezésével egy bázikus, amfifil vegyületet nyertem, azonban az Gram-negatív baktériumok ellen inaktívnak mutatkozott. A teikoplaninból és a teikoplanin A3-1 (TB) pszeudoaglikonból kovalens dimereket állítottam elő kétféle bisz-izotiocianát linker segítségével, kétféle orientációban. A dimerek antibakteriális hatása nem múlta felül az anyavegyületekét.


During my PhD research I carried out semisynthetic modifications on two glycopeptide antibiotics, teicoplanin and vancomycin. The aim of this work was to obtain derivatives with high activity against pathogenic bacteria (predominantly Gram-positive) that are resistant to the parent antibiotics and compounds with antiviral activity. A simple, cost-efficient method was developed for the preparation of the teicoplanin A3-2 pseudoaglycon (TC). Then the attachment of side chains with primarily lipophilic character to the N-terminus of this compound was carried out e.g. by copper(I)-catalyzed azide-alkyne click (CuAAC) reaction, N-sulfonylation and by the formation of guanidino groups. Many of the obtained derivatives displayed significantly higher activity against glycopeptide resistant bacterial strains than the parent antibiotics. Some of the compounds also possessed antiviral (e.g. anti-influenzavirus and anti-coronavirus) activity. By systematically modifying the structure of the side chains both the antibacterial and the antiviral activity proved to be adjustable, moreover the reduction of cytotoxicity was also successful in some instances. The incorporation of an amide group substituted by basic amine functions into the C-terminus by peptide coupling (besides the modification of the N-terminus) led to derivatives with even higher antibacterial activity in some cases. Selective antiviral compounds (without antibacterial activity) have been successfully prepared by attaching the side chains of teicoplanin derivatives with the highest activity against influenzaviruses to the N- or C-terminus of vancomycin aglycon hexapeptide (VAHP). With the aim of achieving activity against Gram-negative bacteria, a basic, amphiphilic compound was obtained by the reacylation and the subsequent aminoethylation of VAHP, however, this derivative proved to be inactive against Gram-negative bacteria. Some covalent dimers have been prepared in two orientations from teicoplanin and its pseudoaglycon, teicoplanin A3-1 using two bis-isothiocyanate linkers. The antibacterial activity of the dimers did not exceed that of the parent compounds.
