Egy gomba specifikus protein foszfatáz vizsgálata

Absztrakt

A Candida albicans egy opportunista humán patogén gomba, melynek orvosi jelentősége jól ismert. A C. albicans-ban található egy gomba specifikus Ser/Thr protein foszfatázt, a CaPpz1, amelynek szerepe van a kation transzportban, sejtfal bioszintézisben, morfológiai változásokban, oxidatív stressz válaszban és a gomba virulenciájában. Munkám során modern „omikai” eszközökkel vizsgáltam a CaPpz1 újabb funkcióit, normál és oxidatív stressz körülmények között. Proteomikai eredményeink megerősítették a CaPpz1 szerepét a fehérje szintézisben, a morfológiai átalakulásban és az oxidatív stressz válaszban. Emellett felismertük a foszfatáz egy eddig ismeretlen funkcióját a biofilm képzésben. A transzkriptomikai vizsgálatok során azt találtuk, hogy a CaPpz1 foszfatáz hiánya és az oxidatív stressz önmagában csak kis vagy közepes hatást gyakorol a génexpresszióra, ezzel szemben a két beavatkozás együttesen markáns transzkripciós változásokat okoz, ami a köztük fennálló erős pozitív interakcióra utal. Azt tapasztaltuk, hogy oxidatív stressz hatására csökken a sejtfelszíni fehérjéket és a citoszólikus riboszómális fehérjéket kódoló gének expressziója, míg a plazmamembránokon keresztüli transzportban, oxidációs-redukciós folyamatokban illetve az RNS metabolizmusban részt vevő gének expressziója megemelkedik a foszfatáz hiányos mutáns törzsben. Ily módon a CaPpz1 számos új funkciójára derítettünk fényt. Leglényegesebb eredményünk az, hogy a patogén gombában a CaPpz1 védő szerepet tölt be az oxidatív stresszel szemben, ami felveti annak a lehetőségét, hogy a CaPpz1 foszfatáz specifikus gátlása megfelelő oxidatív kezeléssel kombinálva, egy új gombellenes terápia alapjául szolgálhat.


Candida albicans is an opportunistic human pathogen of great medical significance. C. albicans contains a novel fungus specific Ser/Thr protein phosphatase, called CaPpz1 which has several important physiological roles such as the regulation of cation homeostasis, cell wall biosynthesis, morphological changes, oxidative stress response, and virulence of the pathogen. In my studies I used modern “omics” technologies to investigate its additional functions under normal conditions ad in oxidative stress. With proteomics we confirmed the involvement of CaPpz1 in protein synthesis, morphological transition as well as in oxidative stress response. Furthermore, we proved a novel role for CaPpz1 in biofilm formation. With transcriptomics we found that the lack of CaPpz1 phosphatase or the oxidative treatment alone have only a small or moderate effect on gene expression, but combination they elicit robust changes in the transcriptome indicating a strong positive interaction between the two conditions. We revealed that the expression of genes coding for cell surface proteins and cytosolic ribosomal proteins were downregulated by oxidative treatment, while the amounts of mRNAs associated with transport processes, oxidoreductase activity, and RNA processing were upregulated in the phosphatase deletion mutant strain. Thus we identified several novel functions for CaPpz1. From these results, we conclude that in C. albicans CaPpz1 plays a protective role against oxidative damage, and suggest that the specific inhibition of the phosphatase combined with a proper oxidative treatment may be applied in a new topical antifungal therapy.
