Az aflatoxin képződés vizsgálata mezőgazdasági termékeken

Az Aspergillus sp. penészgombák az egyik legjelentősebb mezőgazdasági kártevők, amelyek mind szántóföldi, mind raktári körülmények között megjelenhetnek. Főként kukoricában és kukorica tartalmú takarmányokban, olajos magvakban, fűszerekben, szárított gyümölcsökön fordulnak elő. Ebbe a csoportba tartoznak a legtoxikusabb mikotoxint, az aflatoxint termelő Aspergillus flavus és Aspergillus parasiticus gombák. Ezek a gombák a növekedésükhöz, szaporodásukhoz, szekunder metabolit termelésükhöz magasabb hőmérsékletet és magasabb páratartalmat igényelnek, ezért eddig elsősorban trópusi területeken fordultak elő. Azonban manapság a klímaváltozás és a szélsőséges időjárási viszonyok miatt hazánkban is megjelentek, ezért vizsgálatuk kiemelkedő jelentőségű. Munkám során 72 db endemikus Aspergillus sp. izolátumot gyűjtöttem össze morfológiai bélyegek alapján, takarmányokról, illetve alapanyagokról. A 72 db izolátum közül 33 db bizonyult potenciálisan aflatoxinogénnek, a toxin szintéziséért felelős génklaszter 3 génjének (aflR, norA, omtA) multiplex PCR-rel való kimutatása alapján. A fajmeghatározást az ITS régió bizonyos szakaszának (ITS1 és ITS4 primerrel) amplifikálása és szekvenáltatása alapján végeztem. Így valószínűsíthetőleg a 33 db potenciálisan aflatoxint termelő izolátumok közül 22 db bizonyult Aspergillus flavusnak, míg egy Aspergillus tritici, három Aspergillus tritici vagy Aspergillus candidus, és egy Aspergillus cristatus vagy Aspergillus amstelodami volt, de a pontos faji azonosításhoz további PCR alapú vizsgálatok szükségesek. Majd a további vizsgálatokhoz kiválasztottam a 22 db izolátum közül egy toxinogént, amely aflatoxintermelését sikerült detektálni maláta agaron, egy atoxinogént, amely nem termelt aflatoxint maláta agaron és a referencia Aspergillus flavus törzset, amelyek faji meghatározását kalmodulin génszekvencia alapján is elvégeztem, ami alapján szintén Aspergillus flavus fajba sorolhatóak be. A természetes közeg modellezésére kukoricaagart vezettem be. A referenciatörzset eltérő tápközegekben vizsgálva azt tapasztaltam, hogy a kukoricagar esetében elsősorban a magasabb zsírtartalom, valamint a glükóz hozzáadása indukálta az aflatoxintermelést, illetve a malátás agaron volt a legnagyobb mértékű az aflatoxintermelés. Tehát a magas antioxidáns tartalom és az alacsony szervetlen nitrogén tartalom ellenére is volt toxinszintézis, ami a glükózból, illetve a zsírsavakból képződő acetil-koenzim A-val magyarázható. A szklerócium produkció, ami a túlélést biztosítja kedvezőtlen környezeti feltételek mellett, kevésbé volt jellemző a malátás agaron, kukoricaagaron pedig elsősorban a magas zsírtartalom és a szervetlen nitrát vegyületek alkalmazása segítette elő a képződését. Tehát nem tapasztaltam szoros összefüggést a szkleróciumprodukció és a termelt aflatoxin B1 mennyisége között. A referencia Aspergillus flavus törzset különböző spóramennyiséggel leoltva malátás agarra azt tapasztaltam, hogy a legkisebb alkalmazott spórakoncentráció eredményezte a legnagyobb mértékű aflatoxin B1 produkciót, valószínűleg a rendelkezésre álló nagyobb mennyiségű tápanyagforrás miatt. Tehát negatív relációs összefüggés mutatható ki a leoltáshoz alkalmazott spóramennyiség, valamint az aflatoxin B1 produkció között malátás agaron. Stresszvizsgálataimat malátás agaron, valamint kukoricaagaron végeztem. Kiderült, hogy a referencia törzs ozmotikus stresszre pigmentáltság változással, valamint növekedett szklerócium produkcióval reagált, mindkét vizsgált táptalajon, míg a sejtmembránt károsító SDS-sel szemben kis koncentrációban fokozott szklerócium képződést, nagyobb koncentrációban pedig csökkent vegetatív növekedést tapasztaltam. Az izolátumok fele hasonlóképpen reagált a NaCl okozta ozmotikus stresszre kukoricaagaron, mint a referencia törzs, viszont az izolátumok egy része nagyobb, néhány izolátum pedig kisebb rezisztenciát mutatott. Malátás agaron pedig csupán négy izolátumnál figyeltem meg nagyobb mértékű szenzitivitást ozmotikus stresszre, az izolátumok nagy részénél csak nagyobb koncentrációban okozott változást a NaCl a vegetatív növekedésben vagy a spóraképződésben. Az SDS hasonló hatást fejtett ki az izolátumokra, mint a referencia törzsre, kukoricaagaron kettő, míg malátás agaron három izolátum már kisebb SDS koncentrációt is nehezen tolerált, csökkent a vegetatív növekedés, spóraképződés, míg a kukoricaagaron öt, malátás agaron egy izolátum bizonyult rezisztensebbnek SDS-el szemben. Tehát összességében megállapítható, hogy az Aspergillus flavus általánosan magas rezisztenciával rendelkezik a vizsgált stresszfaktorokkal szemben, ami fontos lehet növényvédelmi szempontból. Az atoxinogén jelleget HPLC-MS módszerrel erősítettem meg kukoricára oltott izolátumokban. Kiderült, hogy kilenc aflatoxint nem termelő izolátum nem termel egyik vizsgált szekunder metabolitot sem, így alkalmazhatóak lennének biokontrollként szántóföldeken, a kukorica aflatoxin szennyezettségének csökkentésére. Ehhez viszont a további kutatások folyamatban vannak. A kompetíciós képesség vizsgálata során pedig sikerült bebizonyítani, hogy az alkalmazott atoxinogén Aspergillus flavus izolátum képes a kukorica aflatoxin tartalmának csökkentésére toxinogén Aspergillus flavus izolátum alkalmazása mellett. Így tehát alkalmas lehet szántóföldi körülmények között is a kukorica aflatoxin szennyezettségének mérséklésére. A kutatásom folytatásához megfelelő rezisztenciájú kukorica hibridre volt szükség. Különböző kukoricahibridek Aspergillus flavus-szal szembeni ellenállóképességét vizsgálva sikerült találni ellenálló és érzékeny kukoricahibrideket, illetve találtam olyan hibridet is, amely hasonló rezisztenciával rendelkezett a kiválasztott toxinogén és atoxinogén Aspergillus flavus-szal szemben. Ez a későbbi vizsgálataink szempontjából is nagy jelentőségű.


Aspergillus sp. molds are one of the most significant agricultural pests that may appear in both arable and storage conditions. They mainly occur in maize and maize feeds, oilseeds, spices and dried fruits. This group includes the most toxic mycotoxins, and Aspergillus flavus and Aspergillus parasiticus producing aflatoxin. These fungi require higher temperatures and higher humidity for their growth, reproduction, secondary metabolite production, and have so far mainly occurred in tropical areas. However, these days, due to climate change and extreme weather conditions, they have also appeared in Hungary, so their investigation is of paramount importance. In our work, we collected 72 endemic Aspergillus sp. isolates based on morphological characteristics from feeds and raw materials. Of the 72 isolates, 33 were potentially aflatoxinogens as three genes (aflR, norA, omtA) of the aflatoxin gene cluster were detected by multiplex PCR. The species determination was carried out based on the amplification and sequencing of a certain section of the ITS region (ITS1 and ITS4 primers). Thus, of the 33 potentially aflatoxin-producing isolates, 22 were Aspergillus flavus, while one appeared to be Aspergillus tritici, three Aspergillus tritici or candidus, and one Aspergillus cristatus or amstelodami. For further studies, from the 22 isolates a toxinogen isolate was selected, which aflatoxin B1 production was detected on malt agar, an atoxinogen that did not produce aflatoxin B1 on malt agar and the reference Aspergillus flavus strain were also identified on the basis of calmoduline gene sequence and they were also classified as Aspergillus flavus. I introduced corn agar to model the natural medium. Examining the reference strain in different media, I found that the aflatoxin production in the corn agar was primarily due to the higher fat content and the addition of glucose, but malate agar induced the highest aflatoxin production. So, despite the high antioxidant content and low inorganic nitrogen content, there was mycotoxin synthesis, which can be explained by acetyl coenzyme A from glucose and fatty acids. The sclerotium development, which ensures survival under unfavourable environmental conditions, was less typical of malate agar, and its formation was primarily facilitated by high fat content and the use of inorganic nitrate compounds on corn agar. So, we did not detected a close link between the sclerotium production and the amount of aflatoxin B1 produced. The reference Aspergillus flavus strain, which was extinguished with different spore number, was found to have the lowest spore concentration used resulting in the highest aflatoxin B1 production, probably due to the higher available nutrient source. Therefore, there is a negative relationship between the amount of spore used for inoculation and the aflatoxin B1 production. I tested stress on malate agar and corn agar. The reference strain responded with a change in pigmentation to osmotic stress and increased sclerotium production both on the culture medium tested, compared to cell membrane damaging SDS, which increased sclerotium number in low SDS concentrations and reduced vegetative growth at higher SDS concentrations. Half of the isolates reacted similarly to NaCl-induced osmotic stress on corn agar than the reference strain, but some isolates have increased and some isolates showed less resistance. In only four isolates, I observed greater sensitivity to osmotic stress, and in most isolates, NaCl only caused a change in vegetative growth or spore formation in a higher concentration. SDS had a similar effect on isolates as the reference strain, and two isolates on maize agar, and three isolates on malate agar hardly tolerate low SDS, they suffered decreased vegetative growth, spore formation, while five on corn agar, an isolate of malate agar has been found to be more resistant to SDS. So overally, it can be concluded that Aspergillus flavus has a generally high resistance to the stress factors studied, which may be important from a plant protection point of view. To continue my research, a maize hybrid with sufficient resistance was needed. By examining the resistance of various maize hybrids to Aspergillus flavus, I was able to find resistant and sensitive maize hybrids, as well as a hybrid with similar resistance to the selected toxinogenic and atoxinogenic Aspergillus flavus. This has also a great importance for our future investigations. In the study of the to competition ability, I demonstrated that the atoxinogenic Aspergillus flavus isolate could reduce the aflatoxin content of maize inoculated with toxinogenic Aspergillus flavus isolate. This isolate may therefore be suitable for reducing the contamination of maize with aflatoxin even under arable conditions. Atoxinogenicity was confirmed by HPLC-MS in isolates inoculated on corn. It was found that nine isolates did not produce aflatoxins neither produce any of the secondary metabolites tested and could therefore be used as biocontrol in arable fields to reduce the aflatoxin contamination of maize. To do this, however, further research is ongoing.
