Due to their serious health issues, there is growing interest in developing safer alternatives to gadolinium-based MRI contrast agents (GBCA). Manganese(II) is considered a potential alternative metal due to its essential nature as a micronutrient, and ligands that can chelate manganese have been extensively studied. This research has developed a novel contrast agent (CA) candidate by complexing Mn(II) to form [Mn(3,9-PC2A-6-EOB)], which combines the outstanding properties of 3,9-PC2A and the liver specificity of the EOB moiety. The synthesized Mn(II)-based MRI CA candidate [Mn(3,9-PC2A-6-EOB)] has shown high thermodynamic stability and is highly inert towards Zn(II) transmetallation, indicating its superior stability and inertness compared to clinically approved GBCA, gadobenate. This novel CA candidate has excellent potential for future in-vivo studies and ultimately acts as a safer and more effective liver-specific MRI CA.
