The effect of tankyrase inhibitors on the adipocyte cell differentiation

There is an increasing number of people suffering from obesity and diabetes all around the world increase the number of metabolic disease cases such as type 2 diabetes, cardiovascular disease, and even cancer. Recently, TNKS enzymes were identified to play role in metabolic regulation and energy homeostasis and regulate the translocation of Glut4, thus playing a critical role in glucose metabolism. The project aims to investigate how the inhibition of tankyrase affects the differentiation between white, brown, and beige adipocytes utilizing pharmacological inhibitors. To obtain the results we examine the assessment of tankyrase's role in white adipose tissue differentiation and function. We used 3T3-L1 preadipocite cells (mouse origin) as model for white adipose tissue differentiation. As a result, the TNKS1 gene was knockout from T1 cell lines. In further work, we will use that T1 cell line to generate double KO cells.
