Az emlőrák megelőzése és progressziójának gátlása továbbra is jelentős kihívást jelent, különösen a hormonreceptor-negatív emlőtumorok esetén. A retinoid X receptor (RXR) agonisták hormon receptor-független kemopreventív stratégiát képviselnek, klinikai alkalmazásukat azonban dózisfüggő mellékhatások korlátozzák. Ez indokolttá teszi olyan kombinációs megközelítések keresését, amelyek alacsonyabb hatóanyag-koncentrációk mellett is hatékony biológiai választ képesek kiváltani. Jelen doktori munka központi hipotézise alapján, a monoamin jelátviteli útvonalak és a rexinoid jelátvitel kombinált modulációja új, eddig alulértékelt lehetőséget kínál az emlőrák kemoprevenciójában. Vizsgálataink során igazoltuk, hogy a szerotonin (5-hydroxytryptamine - 5-HT) rendszer nem csupán a központi idegrendszerben, hanem normál emlő epithel sejtekben is funkcionális jelátviteli szereppel bír, és alapvetően befolyásolja a sejtek proliferációját. Eredményeink alapján a monoaminerg GPCR-jelátvitel iránya meghatározó a normál emlő epithel sejtek proliferatív válaszában. A kombinációs megközelítés molekuláris hátterének feltárása során azonosítottuk a Zymogene Granule Protein 16B (ZG16B) gént, mint olyan közös célpontot, amelynek expressziója a rexinoid–monoaminerg kombinációk hatására szuppresszálódik. A ZG16B funkcionális jellemzése során kimutattuk, hogy a fehérje normál emlő epithel és emlőtumor sejtekben egyaránt fokozza a proliferációt, a migrációt és a tumorsejtek 126 transzformációját. A ZG16B sejtmigrációt fokozó hatásának kialakulásához a PI3K/Akt jelátviteli útvonal aktivációja szükséges.
The prevention of breast cancer and the inhibition of its progression remain significant challenges, particularly in the case of hormone receptor-negative breast tumors. Retinoid X receptor (RXR) agonists represent a hormone receptor-independent chemopreventive strategy. However, their clinical application is limited by dose-dependent adverse effects. This necessitates the development of combination approaches capable of eliciting effective biological responses at lower drug concentrations. Based on the central hypothesis of the present doctoral research, the combined modulation of monoaminergic signaling pathways and rexinoid signaling represents a novel opportunity in breast cancer chemoprevention. In our studies, we demonstrated that the serotonin signaling system is not only functional in the central nervous system but also plays an active signaling role in normal mammary epithelial cells, influencing cellular proliferation. Our results indicate that the directionality of monoaminergic GPCR signaling is a key determinant of the proliferative response in normal mammary epithelial cells. We showed that activation of Gs-coupled signaling pathways, as well as inhibition of Gi- and Gq-coupled receptors, results in antiproliferative effects. Among serotonin-mediated effects, signaling through the 5-HT7 receptor plays a prominent role. 127 Investigating the interactions between monoaminergic signaling and rexinoid treatment, we demonstrated that the combination of bexarotene with monoaminergic modulators (paroxetine or carvedilol) exerts a synergistic antiproliferative effect in normal mammary epithelial cells. During the exploration of the molecular mechanisms underlying this synergistic effect, we identified the Zymogene Granule Protein 16B (ZG16B) gene as a common target, whose expression is suppressed by rexinoid–monoaminergic combinations. Functional characterization of ZG16B revealed that the protein promotes proliferation, migration, and EMT in both normal mammary epithelial and breast tumor cells. Furthermore ZG16B treatment promoted tumorigenic transformation of breast cancer cells. The pro- migratory effect of ZG16B requires activation of the PI3K/Akt signaling pathway. Transcriptomic analyses further demonstrated that ZG16B plays a key role in extracellular matrix remodeling and in the regulation of epithelial–mesenchymal transition (EMT), both of which are suppressed by rexinoid–monoaminergic combination treatment. Overall, these findings highlight that targeted modulation of monoaminergic GPCR signaling, particularly involving the 5-HT system, represents a novel and translationally relevant intervention point in breast cancer chemoprevention. The combined modulation of rexinoid and monoaminergic signaling offers the potential for developing low-dose, long-term preventive strategies. The identification of ZG16B as an epigenetically regulated, secreted factor promoting tumor progression opens new perspectives for biomarker development and targeted preventive approaches.
