Tissue transglutaminase, as known as transglutaminase 2 (TG2), is one of the most characterised members of the enzyme family. Recently, our research group recognised novel RNA-binding ability of Transglutaminase 2 (TG2). Later investigations have indicated that other transglutaminases also share this characteristic. TG6 is expressed predominantly in the central nervous system and has been implicated in neuronal development, motor coordination, and certain neurological disorders. The potential RNA-binding function of TG6 remains largely undefined. To perform its in vitro characterisation and analysis of RNA-binding capability, we developed and set up a TG6 expression system that will later enable the generation of potential site-directed TG6 mutants.
