A mitokondriális membránokban mérhető dipólpotenciál nagysága némileg váratlanul a lizoszómákéhoz adódott hasonlónak. Módszerünk hasznos eszköz lehet azon organellumok azonosítására, amelyekben a megváltozott lipidszintekkel járó betegségek során patofiziológiai szempontból jelentős membránbiofizikai változások következnek be.

Characterization of the dipole potential arising from the ordered orientation of molecular dipoles of membrane phospholipids, sterols, and membrane-associated interfacial water molecules has remained difficult due to the lack of measurement methods suitable for investigating living individual cells, as well as the absence of physiologically relevant compounds capable of experimentally lowering its magnitude. Therefore, in the first part of the work presented in this dissertation, we developed and optimized a high-throughput flow cytometric method based on quantifying the shift in the emission spectrum of the dipole-potential-sensitive F66 fluorophore. This method enables the acquisition of quantitative information on the magnitude of the dipole potential from large numbers of individual living cells. The applicability of our method was validated using several cell lines and treatments, as well as through comparison with the most widely accepted method for studying dipole potential. In our experiments, we also demonstrated the dipole-potential-increasing effects of various sterol derivatives, including 6KC, 7DHC, and cholesterol in living cells. Furthermore, by examining different saturated and polyunsaturated fatty acids, we showed that α-linolenic acid, an ω-3 polyunsaturated fatty acid naturally present in human cell membranes under physiological conditions, was capable of reducing the magnitude of the dipole potential, whereas saturated stearic acid and—interestingly—the ω-6 polyunsaturated γ-linolenic acid exerted the opposite effect. The beneficial action of α-linolenic acid was also observable in the presence of cholesterol, counteracting the latter’s effect. The biological relevance of these changes in dipole potential was demonstrated through their impact on the uptake and particularly the endolysosomal escape of the cell-penetrating peptide penetratin. Due to the favorable spectral properties of F66, it can be excellently combined with other fluorophores, including fluorescent intracellular organelle markers. Based on this, in the work presented in the second part of this dissertation, we optimized a method based on confocal microscopy, quantitative image analysis, and emission ratio measurements of the F66 fluorophore, which enables the comparative investigation of the dipole potential of the plasma membrane and the membranes of various intracellular organelles in living individual cells. Using our novel technique, we demonstrated in several markedly different human cell lines that the magnitude of the dipole potential is highest in the plasma membrane, whereas within intracellular membrane systems it decreases progressively “inward” along the secretory/endocytic pathway in the order lysosome > Golgi apparatus > endoplasmic reticulum. Somewhat unexpectedly, the magnitude of the dipole potential in mitochondrial membranes proved to be similar to that of lysosomes. Our method may serve as a useful tool for identifying organelles in which pathophysiologically significant membrane biophysical changes occur during diseases associated with altered lipid levels.
