dc.description.abstract | Sunflower output globally is threatened by Macrophomina phaseolina (Tassi) Goid's charcoal rot. Macrophomina phaseolina can infect several wild and cultivated host species via seed, soil, or stubble. Controlling charcoal rot is challenging since disease infestation depends on several factors. There are few field infestation management options. Biological, cultural, and chemical controls have been used to manage this disease, but an effective strategy is unclear. The PCR was tried and modified at the University of Debrecen, Institute of Plant Protection according to their laboratory needs. This technique used ITS1 and ITS4 primers. A successful PCR method was developed after multiple repetitions and process reviews. The pure DNA was sequenced with the collaboration by Microsynth, Austria. The DNA sequence was then blasted on the NCBI website, yielding a 96.15% match. Macrophomina phaseolina can be detected early with this approach. Accurate diagnosis and early pathogen detection are crucial to plant disease management. It may be promising for managing and detecting this damaging disease. This work may indirectly reduce economic harm from the Macrophomina Phaseolina fungus, enabling sunflower and other crop cultivation in the future. | |
