dc.description.abstract | The current research was established for the examination of cell specific HCN2 expressional changes in response to stretch in intestinal smooth muscle layer. The expression of HCN2 channel was investigated in macrophages and human intestinal smooth muscle cells (hISMC). Cells were treated with either dimethyl sulphoxide (DMSO) or BMS (inhibitor of I kappa B kinase) to evaluate the response in HCN2 expression when NF-kB pro-inflammatory signalling pathway is blocked. HCN2 protein measurements by Western Blot demonstrated that in macrophages the expression of HCN2 was decreased after ECS stretching, while qRT-PCR showed the significantly decreased mRNA level in hISMC in the ECS protocol. Additonally, BMS treated cells showed higher mRNA signal than in cells treated with control solvent in the hISMC. | hu_HU |
