In addition, our research group has identified a distinct immunological milieu, with increased immunological tuning characterizing the sebaceous gland-rich, sebaceous skin regions compared to the gland-poor, dry areas. However, the onset of the development of these immunotopographical differences is currently unknown. We hypothesize that before puberty, our skin is uniform, and region-specific characteristics only emerge during adolescence due to hormonal changes. To prove this, we aimed to investigate topographically different skin areas derived from healthy children and adults. We performed TaqMan low-density array (TLDA) and examined the gene expression levels of several cytokines, TFs, chemokines, AMPs, and transcription factors in the sebaceous (sebaceous gland-rich [SGR)) and dry (gland-poor [GP]) skin regions of healthy children (0-10 years) and adults (18 years and above) (n=8-8). We did not find significant differences in the examined molecules between the skin regions of children, while, in adulthood, both the dry and sebaceous skin regions exhibited increased immune activity, however, this increase was much more pronounced in the sebaceous skin region with significantly higher expression of IL-17 signaling mediators (e.g. IL-17, IL-1β, lipocalin, S100A family members). Since the most prominent differences were found in the components of IL-17 signaling, we also detected and quantified the source of the IL-17 cytokine in these skin specimens by double immunofluorescence staining at the protein level. Our results suggest that the predominant source of IL-17 is non-pathogenic Th17 cells in all sample types, and Th17 cell count is substantially and significantly increased in the sebaceous skin region of adults. We investigated and compared the expressions of epidermal barrier proteins of healthy, sebaceous gland-rich (SGR) skin and AV samples by immunohistochemistry (IHC). Our results showed significant changes in expressions of proteins in samples with AV compared to SGR skin samples, which have roles in corneocyte envelope formation, desmosome organization, corneocyte desquamation, tight junction formation, barrier alarmins antimicrobial proteins. A LOX részvétele a 2-AG degradációjában különös jelentőséggel bírhat, ugyanis a 2-AG oxidációjával bioaktív metabolitok, többek között leukotriének keletkeznek, amelyek közismert gyulladásos mediátorként számos élettani és patológiás folyamatban szerepet játszanak. Eredményeink alapján a legfontosabb következtetéseink: 1. Az Aurora B, valamint a kinázzal kölcsönható fehérjék expressziója szövettani körülmények között jól vizsgálható. 2. Az immunhisztokémiai és FISH eredmények szöveti microarray használatával reprodukálhatóak. 3. Az Aurora B overexpressziója önállóan, csak a kinázt expresszáló tumorsejtek arányának figyelembe vételével nem értelmezhető. Az overexpresszió szövettani körülmények közötti definiálására egy új indexet (AMI index) vezettünk be, mely figyelembe veszi az adott mintára jellemző sejtproliferációs (mitotikus) aktivitást. Megfigyelésünk szerint az analizált limfóma mintákban az Aurora B relatív overexpressziója magasabb mitotikus aktivitással társul. 4. Az overexpresszió hátterében génamplifikációt nem figyeltünk meg. Invazív emlőtumorok és limfóma mintáknál is a kinázt kódoló gén deléciója volt jellemző, mely minden esetben TP53 delécióval társult. Erős kapcsolatot találtunk a TP53 és az AURKB génkópiaszámok között. Emlőtumor mintákban a két gén együttes vesztése erős korrelációt mutatott az aneuszóm sejtpopulációk megjelenésével. 5. A 17-es kromoszóma aneuszómiája a vizsgált betegcsoportok szöveti mintáiban gyakori jelenségnek bizonyult. A minták FISH-sel kimutatható aneuszómiája jól reprezentálja az egyes minták áramlási citometriával meghatározott ploiditását. 6. Az Aurora B fehérje expresszió és a daganatsejtek ploiditása között direkt összefüggést nem találtunk. 7. A vizsgált daganatok közül a B-ALCL-ben figyeltük meg a legmagasabb AMI értékeket, ez magas 17-es kromoszómaszámmal és fokozott mitotikus aktivitással társult. Megfigyeléseink szerint az Aurora B és a malignus sejtklónok keletkezésének és fennmaradásának egy lehetséges magyarázata az alábbi: a kinázfunkció sérülése a CPC komplex nem megfelelő működésén keresztül mitotikus szabályozási zavart eredményez. A kináz fokozott aktivitása egy fontos mitotikus ellenőrzési pont kiesését okozhatja, ezáltal aneuszómiás sejtpopulációk megjelenését eredményezheti. A TP53 deléciója pedig egy fontos apoptotikus útvonal sérülésének következtében valószínűleg a malignus sejtpopulációk túlélésének kedvez. The main goal of this work was to study the effect of Aurora kinase expression on cell ploidy and tumourigenesis. 50 invasive breast cancer, 50 diffuse large B-cell lymphoma and 10 reactive lymph node sample were recruited in the study. Our main findings: 1. The expression of Aurora B and its binding partners can be studied between histological conditions. 2. Immunohistochemistry and FISH are reproducible methods on tissue microarrays. 3. Because of the significant correlation with the overall cell proliferation rate, the overexpression of Aurora B could not be stated on the basis of kinase expressing tumor cell fractions alone. The relative expression of the Aurora B kinase is better reflected by the AMI index which represents the Aurora B expression in relation to the whole proliferative fraction of the tumor. The higher relative Aurora B expression associated with higher mitotic activity in B-cell lymphoma. 4. FISH analysis of the AURKB locus did not show any gains or amplifications in the samples analyzed. On the other hand, we have observed the loss of the gene is breast carcinoma and lymphoma samples as well. A strong correlation was shown between AURKB and TP53 copy numbers: AURKB loss was associated with TP53 deletion in all samples. According to our results on breast carcinoma, losses at 17p13.1 and chromosome 17 aneusomy determined by FISH showed a statistically significant correlation. 5. Our study presents the frequent occurrence of aneusomy chromosome 17 in breast carcinoma and B-cell lymphoma samples. Chromosome 17 aneusomy evaluated by FISH is correlated with aneuploidy studying by flow cytometry. 6. Direct correlation between kinase expression and ploidy could not be shown. 7. The highest Ami values was seen in B-ALCL samples, it was associated with high chromosome 17 copy numbers and mitotic activity. The damaged Aurora B kinase function serve for regulatory deficiencies in the CPC complex leading to mitotic errors, while p53 deficiency helps malignant cells to survive due to insufficient activation of the intrinsic apoptotic pathways. The upregulation of Aurora kinase B function results an error in an important mitotic checkpoint, thus resulting in induction of aneuploid cell populations. These parallel effects finally increase the complexity of mitotic abnormalities and generate aneuploid cell populations.
