dc.description.abstractEurópa-szerte a nagytestű vadfajok általános térhódítása jellemző az elmúlt évtizedekben. Ezek közül is az egyik kiemelkedő faj a vaddisznó, mely a világ 100 leginvazívabb élőlénye közt is helyet kapott kivételes adaptációs képességének hála. Disszertációm ezért a vaddisznó genetikai és populációdinamikai vizsgálatával foglalkozik, mivel hazánk egyik legjelentősebb vadfajáról van szó, melynek genetikai háttere ezidáig nem kapott elég figyelmet. A Kárpát-medencén belül négy országból összesen 486 vaddisznóból gyűjtöttünk mintát, amit egy 13 markert tartalmazó egyedazonosításra használt STR, valamint egy 3 markerből álló, a vaddisznókat a házi sertésektől elválasztó InDel markerszettel vizsgáltunk le. Az eredmények alapján először határoztuk meg a magyarországi alpopulációk genetikai mutatóit, valamint a méretüket, a területi elhelyezkedésüket és az azokat meghatározó tényezőket. Megállapítottuk, hogy két csoport található hazánkban, melyek egy populáció különböző alpopulációi, valamint hogy az elkülönülés egy kisebb, az ország északnyugati részét lefedő és egy nagyobb, a többi területet tartalmazó részre osztja azokat. Továbbá nyomokat találtunk arra vonatkozóan, hogy mind az utolsó jégkorszak, mind az 1800-as évek végén kezdődő vízrendezés hatásai a mai napig érződnek a magyarországi populációban. A populációgenetikai rész lezárásaképp az egyedazonosító markerszettet teszteltük le arra vonatkozóan, hogy valóban képes-e minden egyedet szétválasztani. Az általunk vizsgált 486 vaddisznó összesen 485 különböző genotípusba tartozott, a két megegyező genotípusú egyedre pedig biológiai magyarázat adható. Populációdinamikai vizsgálataink során kimutattuk, hogy bár genetikai beszűkülést okozó esemény a hazai vaddisznóállományban jelenleg nem zajlik, de a múltban erős palacknyak-hatás érte a populációt, mivel szinte az összes vizsgált allélon (SMM: 13/13, TPM: 11/13) szignifikáns heterozigóta hiány mutatkozik. A rokonsági vizsgálatokban a mikroszatellita markerszett alapján 23, a kombinált markerszett alapján pedig 22 testvérpárt találtunk 90%-os valószínűségi szinten. Féltestvéri kapcsolatokat 18-at, illetve 29-et, szülő-utód rokonságot pedig kizárólag a kombinált markerszett eredményei alapján sikerült kimutatni, ahol 2 anya összesen 9 utódját találtuk meg. Disszertációm záró részében egy korábban hústermék-nyomonkövetésre kifejlesztett, a vaddisznót a házi sertésektől elválasztó és vegyes genotípust is kimutatni képes markerszettet teszteltünk a vad populáción a hibridizáció megállapítása céljából. Ebben az esetben 422 vaddisznót tudtunk a vizsgálatba bevonni, mely során először csak a 3 hibridizációs (InDel) markert használtuk fel, majd a hibridizációs markerszettet és az egyedazonosító (STR) markerszettet összevontuk. Eredményeink alapján az InDel markerszett 85,5%-ban a valós eredményeket adja, hamis pozitív eredményt a hibridizáltságra (javítható hiba) 13,41%-ban, míg hamis negatív eredményt 1,09%-ban. Mivel ezt a markerszettet eredetileg elővizsgálatok számára fejlesztettük ki, így ezt az eredményt kifejezetten jónak könyveltük el, ami még a megerősítő vizsgálatokkal együtt is lényeges mennyiségű pénzt és időt spórol meg. A szett megbízhatósága után a pontosságát is levizsgáltuk. Ehhez 23 teljes genomra (11 házi sertés és 12 vaddisznó) illesztettük az általunk használt InDel, valamint a szakirodalomban leggyakrabban előforduló markereket. Az eredmények alapján a mi három markerünkből kettő (W1: 89,13%, W2: 70%) önmagában is megállja a helyét a régebben használt markerekkel szemben (MC1R1 és MC1R2: 36,84%, MC1R3: 21,43%, NR6A1: 80,56%), a három marker kombinációja pedig korábbi vizsgálataink alapján 95% fölötti pontosságot biztosít.
dc.description.abstractIn recent decades the distribution area of large game species has been generally increasing across Europe. One typical example is the wild boar, which is one of the most invasive species in the world due to its superior adaptability. Therefore, my dissertation is written about the genetics and population dynamics of the wild boar, as it is one of the most widespread and important game species in Hungary, but its genetic background has not received enough attention so far. Within the Carpathian Basin we collected samples from a total of 486 wild boars in four countries, which were analyzed with 13 STR markers used for individual identification and 3 InDel markers used for separating wild boars from domestic breeds. Based on the results, we first determined the basic genetic indicators of the Hungarian subpopulations, as well as their size, spatial location and the barriers that separate them. We found that there are two groups in Hungary, which are different subpopulations of a single population, of which the smaller one is located in the northwestern part of the country, and the larger one contains the other areas. Furthermore, we found traces that the effects of both the last ice age and the water management that began in the late 1800s can still be seen in the Hungarian population nowadays. In the last part of the population genetics section, the individual identification marker set was tested to see if it could really separate all individuals. The sample set of 486 wild boars studied belonged to a total of 485 genotypes, and the two individuals with the same genotype can be biologically explained. Our population dynamics studies have shown that although there is currently no evidence of an event causing genetic depletion in wild boars, the population was strongly affected by bottlenecks in the past, as there is a significant deficiency in heterozygosity in almost all examined alleles (SMM: 13/13, TPM: 11/13). In the kinship studies 23 sibling pairs were found at the probability level of 90% based on the microsatellite marker set and 22 sibling pairs based on the combined marker set. Also, respectively 18 and 29 half-sibling pairs were found, but parental-offspring relatedness was found only by the results of the combined marker set, where a total of 9 offspring of 2 mothers were found. In the last part of my dissertation a marker set was tested - which had been previously developed for tracking meat products, that separates wild boar from domestic pigs and can also detect mixed genotypes – in wild population to determine the level of hybridization. In this part we were able to include 422 wild boars in the study. At first, the 3 InDel markers were used, and then this set was combined with the STR set. Based on our results, the InDel marker set gives valid results in 85.5%, a false positive result for hybridization (which can be corrected by later tests) in 13.41% and false negative results in 1.09%. Since this set was originally developed for pre-screening, this result was considered to be exceptionally good for studies, because even with confirmatory testing it saves a significant amount of money and time. To check the accuracy of the set, the InDel and the most commonly used markers were aligned to 23 whole genomes (11 domestic pigs and 12 wild boars). Our results showed that two of our three markers (W1: 89.13%, W2: 70%) can stand out alone from the previously used markers (MC1R1 and MC1R2: 36.84%, MC1R3: 21.43%, NR6A1: 80.56%), and the combination of the three markers provides an accuracy of over 95% based on our previous studies.
